Targeted protein degradation has emerged as one of the most powerful therapeutic modalities in modern drug discovery, offering the ability to eliminate disease-driving proteins that are inaccessible to conventional small-molecule inhibitors. Among the available E3 ligases, cereblon (CRBN) remains the most clinically validated and widely exploited, forming the foundation of both PROTACs and molecular glue (MG) degraders.


However, molecular glue discovery continues to face major bottlenecks, including the lack of rational design strategies and the limited chemical diversity of available E3 ligase binders. These challenges restrict the efficiency of molecular glue discovery and design, driving strong demand for new toolkits for molecular glue screening.
